Peptide Reconstitution Guide

Step-by-step instructions for reconstituting lyophilised research peptides and storing them correctly in in vitro laboratory settings.

Before you begin

  • Allow refrigerated or frozen vials to reach room temperature.
  • Inspect the seal and lyophilised cake for moisture or damage.
  • Prepare bacteriostatic water or sterile water per your SOP.
  • Use clean gloves and a low-particulate work surface.

Calculate diluent volume

Final concentration equals vial mass divided by solvent volume. A 10 mg vial plus 2 mL solvent yields 5 mg/mL. Choose a volume that keeps your planned aliquot in a comfortable pipette range — typically 50–500 µL.

Add diluent correctly

Direct bacteriostatic water slowly down the inside wall of the vial — not onto the lyophilised powder. High-velocity streams cause foaming and mechanical stress. After adding the measured volume, gently swirl or roll the vial until fully dissolved. Avoid vigorous shaking unless your protocol requires it.

Label and verify

  • Label with compound name, concentration, batch number, and date.
  • Compare appearance with the batch Certificate of Analysis.
  • Log the vial in your laboratory inventory system.

Storage after reconstitution

Most reconstituted research peptides are stored at 2–8 °C for short-term use. For longer timelines, aliquot into microcentrifuge tubes and freeze at −20 °C or −80 °C to limit freeze-thaw damage. Keep vials upright, protected from light, and away from repeated room-temperature exposure.

Lyophilised material not yet reconstituted is best kept at −20 °C for extended storage, or at 2–8 °C for near-term use.
