How to Reconstitute Lyophilised Peptides: A Laboratory Guide
Step-by-step guidance on adding bacteriostatic water, achieving the right concentration, and preparing samples for in vitro work.
Lyophilised (freeze-dried) peptides arrive as a light powder in sealed vials. Before they can be used in cell culture, binding assays, or other in vitro models, they must be reconstituted in a suitable diluent. Doing this carefully preserves peptide integrity and makes downstream measurement more reliable.
Before you begin
- Allow the vial to reach room temperature if it was refrigerated or frozen.
- Inspect the lyophilised cake for moisture ingress or damage to the seal.
- Prepare bacteriostatic water (0.9% benzyl alcohol) or sterile water as required by your SOP.
- Use insulin syringes or precision pipettes calibrated for small volumes.
- Work in a clean area; gloves and low-particulate surfaces reduce contamination risk.
Adding diluent without damaging the peptide
Direct the diluent slowly down the inside wall of the vial — not onto the lyophilised material itself. High-velocity streams can cause foaming and mechanical stress on the peptide matrix. After adding the calculated volume, gently swirl or roll the vial. Avoid vigorous shaking unless your protocol specifically requires it.
Calculating volume

After reconstitution
- Allow the solution to fully dissolve — most peptides clear within a few minutes.
- Label the vial with date, concentration, and batch number.
- Store according to compound-specific stability data (often 2–8 °C short term).
- Aliquot if you will freeze-thaw repeatedly; each cycle can reduce activity.
