Peptide Stability After Reconstitution: What Researchers Should Know

Factors that influence how long reconstituted peptides remain usable — pH, temperature, diluent choice, and handling.

Speculation about peptide stability abounds in online forums — sometimes claiming compounds become useless within hours of reconstitution. Published analytical work and standard laboratory practice tell a more nuanced story. Stability depends on the specific sequence, formulation, storage temperature, and how often the vial is accessed.

Diluent selection

Bacteriostatic water contains 0.9% benzyl alcohol, which suppresses bacterial growth in multi-dose vials used over days or weeks. Sterile water without preservative is appropriate for single-use aliquots frozen immediately. Some peptides require dilute acetic acid or other buffers — check literature and COA notes before deviating from standard water-based reconstitution.

Temperature and time

  • 2–8 °C refrigeration is standard for many reconstituted research peptides.
  • −20 °C or −80 °C extends life for aliquots not needed imminently.
  • Room-temperature exposure during bench work should be minimised and logged.
  • Light-sensitive sequences benefit from amber tubes or foil wrapping.

MOTS-c and other stability-sensitive sequences

Mitochondrial-derived peptides such as MOTS-c receive particular attention in stability discussions. Analytical monitoring — not anecdote — is the reliable approach. If your assay is sensitive to degradation products, schedule fresh reconstitution before critical runs and retain a retained sample for comparison testing.
